Frequently Asked Questions

1. Do you offer a trial version of the OTMIAS software?

Technically no. However, every now and then we will have a limited time offer where we post a coupon on our website that enables the user to get free one year license of fully functional OTMIAS software. Check the “special Offers” tab in the “Store” pull down menu. This offer usually comes with downloadable image files that you can use to practice using OTMIAS in conjunction with the corresponding video tutorials available on this website.

2. How many images do I have to capture and analyze from each test slide?

Sections of O-STANDARD blocks for proliferative activity or apoptosis show heterogeneity in the percentage of positive cells in different areas of the  sections. For there two tests  (proliferative activity/SPF and apoptosis) you should take at least two separate images each from different area of the stained section to include an area with high percentage of positive cells and area of lower percentage of positive cells. For biomarker analysis O-STANDARDS , one image of the positive standard and one of the negative standard is sufficient. For your test slide, it is up to you to decide whether you want to focus on one area of your slide or  get an average read of a larger area of the test slide.

3. Why did you stop selling cytologic preparations as standards for DNA ploidy analysis?

Any person using OTMIAS® for DNA ploidy analysis should be able to produce his or her own standard slides for this assay. If working with cells, you can use any cell line that is known to be diploid. You can also make touch preparations of freshly cut normal/reactive human (or mouse or rat, .. etc) tonsils, if you have access to such fresh specimens. Always use controls/standards from the same species as your test sample, and prepare your standard slides in exactly the same way as you prepare your test sample for the assay.

4. Do I have to have administrator privileges to use OTMIAS?

You have to have administrator privileges to install the software and to download reference values for OSTANDARDS if they are not already installed in the software copy you are using.

5.  What image formats can I use with OTMIAS?

Only TIFF images can be used. Before you start OTMIAS, please check the image files and make sure they all end with .TIFF even though you are sure they are TIFF files.  If the image name does not end with .TIFF then please add it. For example, if your image file name is “test” then make it “test.TIFF”.

6. Can I use OTMIAS on more than one computer?

Yes. Your OTMIAS license allows you now to install and activate two copies on  computer that you own. Please note that you have to activate the software every time you install it.

7. Can I use my own standard cell lines for biomarker that analysis instead of purchasing O-STANDARDS?

Yes.  You can make your own standard positive and negative cell lines for any biomarker you like. The potential disadvantage with this is that others using OTMIAS may not be able to  correlate their results with yours if each uses different standards. If you use your own standard slides with OTMIAS, you should enter “201-GB” for generic biomarker when you are prompted to do so.

8. What is the best microscope objective magnification for capturing TIFF images to use with OTMIAS?

Capture images using only 20X or 40X microscope objective. DO not use oil.
