DNA ploidy cell cycle analysis

OTMIAS® can be used for DNA ploidy cell cycle analysis on Feulgen-stained sections or cytologic preparations of intact cells. External diploid standard slides must be used when the test sample does not include identifiable internal normal diploid cells, like when working with cell lines for example.  If such cells are not present, then lymphocytes in a section or touch/cytologic preparation of normal tonsil (from the same species as the test sample) can be used.

 

In the figure to the right, we show an example of DNA ploidy analysis on a touch preparation of human tonsil fixed in formalin and stained with the Feulgen method. OTMIAS® handles diploid control cells and sample cells separately. The upper histogram (green) represents the normal diploid control (lymphocytes) with only 6 lymphocytes used (using of up to 100 lymphocytes instead of just 6 gives the same result). The bottom histogram (red) represents analysis on a similarly processed touch preparation from a different tonsil using 101 test cells . This is to show the reliability of the test.

 

The boundaries for G0/G1, S, and G2/M are automatically set by OTMIAS® based on the diploid standard. Detailed results with the number and percent of cells in each phase of the cell cycle, including cells >5C and  >9C, and the C.V. values, are given in a table.
